内洋地黄素与脑血管疾病

内洋地黄素又称内源性地高辛样物质(endogenous digitalis-like substance, EDLS)、内源性地高辛样因子(endogenous digitalis-like factor, EDLF)或内源性哇巴因样物质(endogenous ouabain-like substance, EOLS),是体内产生和分泌的一种具有洋地黄样生物活性的物质,主要分布于下丘脑和肾上腺,具有利尿利钠、缩血管和强心作用。研究证明,细胞膜Na+,K+-ATP酶的α-亚基是洋地黄的受体,内洋地黄素是洋地黄受体的内源性介质,能显著抑制Na+,K+-ATP酶活性。近年来的研究发现,患有脑血管疾病的人和动物,体内内洋地黄素的水平发生改变,其含变化与脑血管痉挛和意识改变程度相关。本文对内洋地黄素的生理作用及其与脑血管病的关系作一介绍,并探讨其拮抗剂可能在脑血管疾病中的应用前景。
1 内洋地黄素的分泌与生物学活性
内洋地黄素在体内分布广泛,存在于脑、心、肝、肺、肾、肌肉、肾上腺和胎盘等多种组织,以及脑脊液、血浆和尿液中。大鼠下丘脑含量最高,约为(101±13) pg/mg,心脏含量为(52±7) fmol/pg,豚鼠心脏含量最丰富,红细胞内也含有。
