(毕业论文 字数:5160 页数:6)摘 要: 用勒杜鹃茎段为外植体,研究不同的外植体的接种方式与不同激素浓度配比对无菌系建立的影响,在此基础上进一步研究诱导勒杜鹃无根苗试管开花的外源激素浓度组合、蔗糖与琼脂浓度的组合,以及光照、温度等培养条件。结果表明:将茎段纵切,除去1/3组织水平接于MS+6-BA0.05mg/L培养基中,增殖系数最高,达到4.06;自然光照下, pH值=5.6,蔗糖浓度为3.0%,琼脂浓度为0.6%,无根苗在MS+6-BA0.05mg/L培养基中诱导试管开花时,开花诱导率最高,达到45.83%。 关键词:勒杜鹃,无菌系,试管开花 Study on Establishing of Clone and In vitro Flowering Induction of Bougainvillea spectabilis Willd. Abstract: The sterilized indefinite shoots induced from the axillary’s buds of Bougainvillea spectabilis Willd. stem were used as the material, the effects of varies treatment on establish of germfree system and flower inducing such as the mode of inoculation of explants, external hormone, nutrition environmental condition and so on were studied respectively. These results were indicated that, slit an incision vaccinate in MS+6-BA0.05mg/L, propagate is the highest, about 4.06. In MS+6-BA0.05mg/L, under natural lighting and temperature, value of pH about 5.6, the concentration of sucrose and the agar is 3.0% and 0.6% separately, dispersion of in vitro flowering is 45.83%.
