[分子生物学] RNA干扰（RNAi）的现状与展望

在mRNA水平上寻找一种既方便又可靠的方法敲除基因表达是近15年来分子生物学家梦寐以求的。为了获得功能缺失的细胞或有机体，各种各样的方法包括反义序列、核酶、嵌合的寡聚核苷酸都试验过，但这些分子的设计均基于试验，难免有错误，有赖于目的基因的性质，结果难以预测，因此往往只产生微弱的抑制作用 (Braasch,Corey,2002).
